CRISPR–Cas systems are RNA-guided immune systems in bacteria and archaea that have been repurposed for programmable nucleic acid targeting and editing. Despite their broad utility, widely used CRISPR effectors remain constrained by size, targeting requirements, variable activity and specificity, and potential immunogenicity and cellular toxicity, limiting their applications in diagnostics and therapeutics.

Our laboratory explores the natural diversity of CRISPR–Cas systems to identify new effectors with distinct and advantageous properties. Through computational mining, we discover previously uncharacterized systems and investigate their molecular properties and mechanisms of action through biochemical, structural, and functional studies. We then apply AI-assisted protein engineering, including protein and nucleic acid language models, to optimize promising candidates and develop them into diagnostic and therapeutic tools. Our current research focuses on sensitive and multiplexed RNA detection, sequential RNA recording in single cells, efficient large-fragment DNA knock-in and deletion, and precise RNA editing and knockdown. Ultimately, we aim to expand the CRISPR toolbox and enable more precise and tailored approaches to diagnostics and therapeutics.
